Ca²⁺ — the preeminent intracellular second messenger. In the heart, a 10-fold cytosolic Ca²⁺ transient (100 nM → 1 µM during systole) triggers troponin C and every contraction via CICR from the SR. β1-AR/PKA signaling and SERCA2a tune the Ca²⁺ transient.
