IL-10 is the master anti-inflammatory cytokine; IL10 gene chr1q32.1; homodimer; IL-10Rα/IL-10Rβ → JAK1/TYK2 → STAT3 → suppresses NF-κB in macrophages, reducing TNF-α, IL-1β, IL-6, IL-12. Treg-derived IL-10 protects against GvHD; IL-10R mutations cause very-early-onset IBD.
