Small GTPase and proto-oncogene; mutated in ~25% of all human cancers (PDAC ~90%, CRC ~45%, NSCLC ~30%). Oncogenic mutations (G12D, G12V, G12C) lock KRAS in GTP-bound active state, constitutively activating RAF-MEK-ERK and PI3K-Akt-mTOR. KRAS G12C now targetable by sotorasib.
